急性缺血对豚鼠心室肌细胞L型钙通道电流的影响

Effect of Acute Ischemia on L-type Calcium Channal Current in Isolated Guinea Pig Ventricular Myocytes

目的 观察急性缺血对豚鼠心室肌细胞 L型钙通道电流 (ICa- L)的影响。 方法 采用膜片钳全细胞记录方法 ,分别于模拟急性缺血后 0 ,5 ,10 ,15和 2 0 m in记录 ICa- L。 结果 模拟急性缺血状态后豚鼠心室肌细胞ICa- L明显受抑 ,L钙电流峰值最大值较对照组下降 2 5 .6 5 %± 4 .13% (P<0 .0 5 ) ;但其最大活性电压和 ICa- L的 I- V曲线形态无明显改变。 结论 模拟急性缺血状态可明显抑制豚鼠心室肌细胞 L 型钙通道电流 ,但不影响最大活性电压和 ICa- L的 I-

Objective\ To investigete the effect of acute ischemia on the L\|type Ca\+\{2+\} channel current(I\-\{Ca\|L\}) in guinea pig ventricular myocytes.\ Methods\ Using the technique of whole cell patch clamp to determine basaline and 5, 10, 15 and 20 min after acute myocardial ischemia for detection of the effect of the L\|type Ca\+\{2+\} channel current in isolated guinea pig ventricular myocytes.\ Results\ The peaks of I\-\{Ca\|L\} were decreased obviously by 25 65%±4 29% compared with the control group during acute ischemia(P<0 05); but there was no any significant changes on maxium active voltage and I\-\{Ca\|L\} I\|V relation curve.\ Conclusion\ The I\-\{Ca\|L\} in isolated guinea pig ventricular myocytes is inhibited prominiently by acute \{ischemia\}; and there is no influence to maxium active voltage and I\-\{Ca\|L\} I\|V relation curve.\;
