Synthetic improvement and animal experiment of 6- ^( 18)F-DOPA

摘要 目的 研究 6 1 8F 多巴 (DOPA)的合成改进及其在大鼠体内和偏侧帕金森病 (PD)大鼠模型脑内生物分布。方法 以 6 硝基胡椒醛为前体 ,经亲核氟化、二碘硅烷在柱还原碘化、手性相转移催化烷基化及水解 4步反应合成 6 1 8F DOPA ,测定大鼠体内和偏侧PD大鼠模型脑内 6 1 8F DOPA生物分布。结果  6 1 8F DOPA总放化产率为 5 %~ 18% (未经时间衰减校正 ) ,总合成时间 <110min,放化纯度 >98% ,对映纯度 >97%。正常大鼠肾脏、血液、纹状体和海马对 6 1 8F DOPA摄取较高 ,但前两者对 6 1 8F DOPA清除快 ,而后两者对 6 1 8F DOPA滞留时间长 ,且具有较高纹状体 皮质和纹状体 小脑放射性比值。与假手术对照组和偏侧PD大鼠模型未损毁侧 (左侧 )比较 ,偏侧PD大鼠模型损毁侧(右侧 )纹状体对 6 1 8F DOPA摄取显著下降 ,且具有较低纹状体 皮质和纹状体 小脑放射性比值 (P <0 0 5 )。结论 提供了简便实用的 6 1 8F DOPA合成方法 ,制备的 6 1 8F DOPA可用于动物及PD患者PET显像。 Objective Tostudysyntheticimprovementandbiodistributionof 6 1 8F DOPAinnormalrats andhemi Parkinsonismrats .Methods 6 1 8F DOPAwassynthesizedfromthestartingmaterial 6 nitropiperonal viamulti stepreactionincludingthenucleophilicfluorination ,reductiveiodinationwithdiiodosilaneonSep Pak column ,chiralcatalyticphase transferalkylation ,andhydrolysisreaction .Biodistributionof6 1 8F DOPAinnor malratsandthebrainofhemi Parkinsonismratswasdetermined .Results Thetotaltimeofsynthesiswasless than 110min ,thetotaluncorrectedradiochemicalyieldfrompotassium 6 1 8F DOPAwas5 %~ 18% ,andtheen antiomericpurityandradiochemicalpuritywereabove 97%and 98% ,respectively .Highuptakeinthekidney, blood ,striatum ,andhippocampus,rapidbloodclearanceinthekidneyandblood ,longretainingtimeandhigh striatum cerebellumandstriatum cortex 6 1 8F DOPAuptakeratiowerefoundinnormalrats.Comparedwiththe intactsideofhemi Parkinsonismratsandpseudo operatedgroup ,6 1 8F DOPAuptakeandstriatum cerebellumand striatum cortex 6 1 8F DOPAuptakeratioreducedsignificantlyinthelesionedsideofhemi Parkinsonismrats (P < 0 .0 5 ) .Conclusions Asimpleandpracticaltechniqueisprovidedfortheradiochemicalsynthesisof6 1 8F DOPA . Thesynthetic 6 1 8F DOPAisallowedtobeusedtostudytheanimalandParkinson’sdiseasewithPETimaging.
