异莲心碱对心血管功能及压力超负荷型左心室肥大的作用 被引量:22

摘要 在离体心肌标本、毁脊髓大鼠及腹主动脉部分结扎所致心室肥大大鼠模型上 ,观察异莲心碱 (IL)对心血管功能及压力超负荷型左心室肥大 (L VH)的作用。结果发现 :IL 可剂量依赖性的抑制左心房收缩力 (Fc)及右心房自搏频率 (Freq) ,并使 Ca2 +量效曲线右移 ,最大反应降低 ,其 p D2 ’ =4 .7;它对抗苯肾上腺素 (PE)升高毁脊髓大鼠血压的作用 ,IL 显著削弱腹主动脉部分狭窄所致的收缩压 (SAP)、舒张压 (L VSP)、左室舒张末压 (L VEDP)及压力变化最大速率 (- dp/dtmax)升高 ,减轻心脏重量 ,降低心脏羟脯氨酸含量。结果提示 :IL 可减轻 L VH,其抗 L VH作用可能与非竞争性的抑制外 Ca2 + 内流 ,阻断α1 In order to investigate the effects of isoliensinine(IL) on cardiovascular function and left ventricular hypertrophy (LVH), a rat model of LVH was established by destroying spinal cord and partial ligation of abdominal aorta. The action of IL (10 or 20 mg/kg every day for 6 weeks) on contractile force (Fc), pacemaker frequency and Ca 2+ dose response curve were examined in isolated left or right atria. Its effect on systolic arterial pressure (SAP) and diastolic arterial pressure (DAP) were observed in pithed rat pretreated with PE. The hemodynamics, whole heart weight (WHW)/body weight (BW), left ventricular weight (LVW)/body weight (BW), and hydroxyproline (Hyp) content were measured. It was found that IL produced a dose dependent negative inotropic with IC 50 value of 39.8 μmol/L and a chronotropic effect, shifted Ca 2+ dose response curve to the right, decrease the maximal response with the pD 2 ' value of 4.66. IL also abolished the increasing effect of PE on SAP and DAP. In the rats with LVH, IL decreased SAP, LVSP, LVEDP, and increased -dp/dtmax. Moreover, it reduced the WHW/BW, LVW/BW, and Hyp content ( P <0 05 or P <0 01). It was concluded that IL blocked Ca 2+ influx and antagonized the function of α 1 adrenergic receptor, which might responsible for its protecting heart from LVH.
