摘要
显微注射法建立转 bcl- xl基因小鼠 ,将 PCR及 Southern- blot检测阳性的小鼠与正常鼠交配并传代 ,然后检测子代中外源基因的遗传情况。结果发现 :10号小鼠传代过程中 PCR阳性率保持稳定 ,符合孟德尔遗传规律 ,外源基因能够稳定遗传。 6号小鼠 PCR阳性率呈轻微下降趋势 ,但能够保持相对稳定遗传。 13号和 5号小鼠 PCR阳性率均呈现明显的下降趋势。该二小鼠存在外源基因遗传丢失现象。结果提示
Human bcl xl transgenic mice were developed by microinjection and confirmed by PCR and Southern blot in the founders. The transgenic mice lines were established by mating transgenic mice with wild type mice. The offsprings were identified by PCR and Southern blot to detect the transmissibility of foreign gene. It was found that the PCR positive rate of No.10 mouse kept stable during passage. The PCR positive rate of No.6 mouse presented the tendency of a slight decrease during passage. The PCR positive rate of No.13 and No.5 mice showed the tendency of a notable decrease during passage. It was suggested that the foreign gene in these two mice had been partly lost during transmissibility. It could be concluded that the mouse line in which the foreign gene can be stably reproduced and transmited was obtained.
