摘要
目的 研究胃癌、癌旁组织中线粒体膜蛋白 (Apo2 .7) ,Fas和细胞凋亡关键性调控基因Bcl 2蛋白及相互关系 ,进一步探讨胃癌发病可能的分子机制。方法 胃癌患者 45例 ,术中取胃癌组织 ,癌旁组织各 1块 ,用流式细胞仪定量检测Apo2 .7及Fas、Bcl 2基因蛋白的表达。结果 胃癌组织的Apo2 .7表达 (1 2 .89± 3 .57) %显著低于癌旁组织(1 9.2 8± 6 .1 6) % (P <0 0 1 ) ,而Bcl 2蛋白在胃癌组织中的表达 (1 5 .96± 5 .0 8) %显著高于癌旁组织 (1 0 46± 2 .94) %(P <0 0 0 1 ) ,Fas蛋白在两者中差异无显著性 (P >0 0 5) ;Apo2 .7表达与分化、浸润程度有关 (P <0 0 5) ,Fas表达与浸润程度、TNM分期有关 (P <0 0 5) ;Fas与Bcl 2、Fas与Apo2 .7表达之间均无显著相关性 (P >0 0 5) ,而Apo2 .7与Bcl 2表达之间存在显著相关性 (P <0 0 5)。结论 在胃癌的发生发展过程中 ,细胞凋亡调控异常起着重要作用 ;Fas、Bcl 2及Apo2 .
Objective To investigate the expression and mutual relationship of apoptosis regulating gene in gastric carcinoma and paracancerous tissues and its moecular mechanism in the course and development of gastric cancer .Methods 45 cases of gastric carcinoma patients underwent surgical resection .Samples were taken from tumor site and Paracancerous tissues.Flow cytometry was used to detect the expression of Bcl 2,Fas Proteins and mitochondrial membrane protein(Apo2.7).Results The expression of Apo2.7 in gastric carcinoma (12.89±3.57)% were significantly lower than that in Paracancerous tissues(19.28±6.16)% ( P <0 01),whereas the positive rate of Bcl 2 gene expression (15.96±5.08)% in gastric carcinoma were significantly higher than that in Paracancerous tissues (10 46±2.94)% ( P <0 001).There were no significant difference( P >0 05)in experssion of Fas in gastric cancer and paracancerous. Significant between Apo2.7 and grade of differentiation and invasion was observed ( P <0 05).The expression of Fas was significantly associated with the grade of invasion and TNM( P <0 05). there were no significant correlation between Fas and Bcl 2,Fas and Apo2.7 ,but significant correlation between the expression of Apo2.7 and Bcl 2 in gastic cancer was observed( P <0 05). Conclusion The abnormal expression of apoptosis may play an important role in the pathogenesis of gastric carcinoma; Fas,Bcl 2 and apo2.7 pay both independent and synergetic role in the course and development of gastric carcer.
