摘要
基因工程酵母菌 Y8经发酵 ,硫酸铵分级沉淀 ,凝胶过滤层析及阴离子交换层析分离纯化 ,获得了电泳纯的植酸酶 (Phyt A)。此酶的分子量约为 69.5 k D,p I为 4 .5 ,最适 p H为 2 .4 ,最适温度为 65℃。与原始出发菌株相比 ,其最适 p H值及等电点相似 ,但分子量增大 。
Geneticengineering barm Y8 was fermented and deposited by (NH 4) 2SO 4, and then the phytase from Y8 was purified over Sephadex G-100 gel filtration resin and anion exchange chromatography DEAE-SepharoseCL-6B to homogeneous product (PhytA). Its molecular weight is 72KD, isoelectric point (pI) is 4.5, optimum pH is 2.4, optimum temperature is 65℃. Compared with the original strain, pH suitability and pI are similar, but molecular weight is bigger and thermostability has increased remarkably.
