摘要
目的 观察羧甲基葡聚糖 (carboxymethyl- β - 1,3 -glucan ,CMG)对脓毒症的防治作用。 方法 采用创伤合并内毒素血症模型 ,观察CMG对小鼠存活率及其肺损伤的影响。体外观察CMG对小鼠肺泡巨噬细胞 (alveolarmacrophages ,AM)清道夫受体 (SR)表达及细胞吞噬功能的影响。 结果 CMG可提高模型小鼠 48h存活率 (P <0 .0 1) ,减轻肺组织炎性损伤 ,抑制内毒素(LPS)对SR表达的下调 (P <0 .0 5和P <0 .0 1) ,增强AM对金黄色葡萄球菌的吞噬功能 ,SR单抗2F8可抑制该吞噬过程。 结论 CMG可通过增加SR介导AM对LPS的清除 ,增强宿主非特异性防御功能。CMG可能是有效治疗脓毒症的免疫调理剂之一。
Objective To investigate the therapeutic effect of carboxymethyl-β-1,3-glucan(CMG) on sepsis in mice. Methods The model mice with traumatic endotoxemia were used to determine the survival and histological effect of carboxymethyl-β-1,3-glucan (CMG) on the lungs. The effect of CMG on the expression of scanvenger receptor (SR) and the phagocytosis of alveolar macrophages (AM) was also investigated in vitro. Results CMG could improve the 48th hour survival rate of the mice with traumatic endotoxemia, mitigate their lung inflammatory injury of pulmonary tissues, inhibit the down-regulation of SR stimulated by LPS (P< 0.05,P< 0.01) and increase the AM phagocytosis of staphylococcus aureus which could be blocked by anti-SR monoclonal antibody, 2F8. Conclusions CMG can increase LPS clearance of AM induced by SR, strengthen hosts' nonspecific defensive function. So CMG may be an effective immunomodulator in treatment of sepsis.
