摘要
目的 探讨基质金属蛋白酶 MMP-2和 MMP-9对人脑胶质瘤侵袭微生态系统的影响。方法 应用透射电镜观察脑胶质瘤侵袭微生态系统中血瘤屏障的超微结构改变 ;用免疫组织化学 S-P法检测 MMP-2和 MMP-9在胶质瘤中的表达情况。并与透射电镜观察结果进行对照比较。结果 (1 )电镜下可见瘤组织中多数微血管基膜的胶质膜侧出现大小不等、多少不一的虫蚀样空洞或血管周围间隙扩大 ;有的血管内皮细胞呈不同程度的增生 ,以出芽方式生成新的肿瘤性血管伴血浆渗出。 (2 )在胶质瘤瘤体内的基膜内皮细胞及瘤细胞可见 MMP-2和 MMP-9表达 ,而在正常脑组织中的血管基膜内皮细胞只见 MMP-2表达。瘤体组织中 MMP-2染色阳性强者其相应的电镜下观察见基膜上虫蚀样空洞明显 ,而 MMP-9染色阳性强者血管外周围间隙扩大明显。结论 MMP-2、MMP-9与人脑胶质瘤侵袭微生态系统中血瘤屏障的改造重建有关。
Objective To explore the influence of MMP 2 and MMP 9 on invasive microecosystem of human brain glioma. Methods The ultrastructure of blood tumor barrier in gliomas was observed by transmission electronic microscope. The expressions of MMP 2 and MMP 9 were detected immunohistochemicaly in the ultrastructure. Results (1)The wormhole like caverns of irregular size and unequal or the enlarged per vascular space in each case were observed on basement membane (BM)near the side of glio membrane.The endothelial cells of microvessels shawed marked proliferation together with tumoral granulation protruded from the deficiency BM by means of budding with plasma protein out of vessel. (2) Both the endothlial cell and tumor cell in tumor entity were stained positively for MMP 2 and MMP 9, where the endothelial cells of normal brain tissue were only positive for MMP 2.Where the stronger MMP 2 expressed,and the bigger the mote eaten cavities were,and the stronger MMP 9 expressed, the larger per vascular space were.Conclusions The overexpression of MMP 2 and MMP 9 may be correlated with the remodelling of blood tumor barrier in human brain glioma.
