明胶酶、IV型胶原、CD44v6在结直肠癌中的表达

摘要 目的 :探讨明胶酶在结直肠癌中的表达及其与IV型胶原和CD44v6在结直肠癌侵袭转移过程中的关系。方法 :采用明胶酶谱和S -P免疫组化对 5 0例结直肠癌癌组织和相应正常组织中的明胶酶、IV型胶原和CD44v6进行检测。结果 :明胶酶在结直肠癌中的表达水平显著高于正常组织 (P <0 0 5 ) ;明胶酶与结直肠癌的Duke’s分期呈同步的正相关 (P<0 0 5 ) ;基底膜中IV型胶原表达减少 ,明胶酶表达显著增加 (P <0 0 5 ) ;明胶酶在CD44v6强阳性组中表达量升高显著(P <0 0 5 )。结论 :明胶酶与结直肠癌的侵袭转移性有着密切的关系 ,不仅在癌细胞的酶解 ,而且在癌细胞的粘附。 Objective To explor the expression of gelatinase,type IV collagen and CD44v6 in colorectal carcinoma and the relationship between gelatinase and type IV collagen,CD44v6 in the process of invasion and metastasis of colorectal carcinoma.Methods Gel zymography and S-P immunohistochemistry were used to examine the expression of gelatinase and type IV collagen,CD44v6 in 50 cases of colorectal carcinoma and the normal tissue of the same samples.Results The contents of gelatinase in colorectal carcinoma were higher than those in normal tissue(P<0.05).The expression of gelatinase was highly related to Duke's stage of colorectal carcinoma(P<0.05).With the high expression of gelatinase,the expression of IV type collagen in basement membrane was low.The expression of CD44v6 in strongly positive group was higher than that in the negative and weakly positive groups(P<0 05).Conclusion There was close relationship between gelatinase and biological action of invasion and metastasis of colorectal carcinoma.Gelatinase not only plays the important role in the process of zymolysis,but exerts some function in the process of adhesion and translation of colorectal carcinoma cell.
