摘要
目的 :筛查胃癌组织中AGO基因突变 ,检测不同肿瘤细胞系中AGO基因的表达情况。方法 :采用聚合酶链反应及变性高效液相色谱法 ,对 2 2例胃癌组织标本中AGO基因 β型全部外显子进行突变筛查 ;应用逆转录 聚合酶链反应法检测多种不同肿瘤细胞系中AGO基因α和 β型转录本的表达情况。 结果 :在所检全部胃癌组织中均未见AGO基因突变 ;AGO基因α和 β型转录本在所有不同肿瘤细胞系中均有不同程度的表达。结论 :AGO基因可能与胃癌的发生和发展无关 ,AGO基因表达缺失在肿瘤细胞中少见。
Objective: Our aims were to screen the AGO gene for mutations in primary gastric carcinomas and to detect the expression of AGO gene in cancer cell lines. Methods: Individual exons of the AGO gene were first amplified by using polymerase chain reaction (PCR) and then subjected to denatured high pressure liquid chromato graphy (DHPLC). For mutation screening, DNA from 22 primary gastric carcinomas were analysed by using PCR DHPLC. The α and β transcripts of the AGO gene were examined by using reverse transcription PCR (RT PCR) in multiple cancer cell lines of different origin. Results: No mutation in regions of the AGO gene screened was revealed in the 22 primary tumor tissues and all the cell lines showed expressions of the two isoforms of the AGO transcripts. Conclusion: Mutations in the AGO gene might not be related to the process of gastric carcinogenesis, and absence of an AGO transcript could be rare in cancer cells.
