Fas和Fas-L在醋氨酚中毒致大鼠肝损伤中的表达

摘要 目的 :探讨醋氨酚中毒大鼠肝组织中Fas和Fas 配体 (Fas L)的表达及其与醋氨酚药物剂量之间的关系。方法 :采用免疫组化方法对醋氨酚中毒大鼠肝组织中Fas和Fas L的表达进行检测。结果 :在绝大多数的大鼠肝组织中均有Fas和Fas L的表达 ,阳性率均为 93 .3 %。醋氨酚中毒剂量与Fas和Fas L的表达之间关系密切(P <0 .0 1) ,醋氨酚剂量越大 ,Fas和Fas L表达越强 ,呈显著正相关 (r =0 .42 5 / 0 .461)。部分阳性肝细胞发现同时有Fas和Fas L的表达 ,但并无淋巴细胞浸润。结论 :醋氨酚介导的Fas和Fas L表达在肝细胞凋亡和坏死中起重要作用 ,肝细胞的直接自杀或自分泌及旁分泌杀伤机制 ,可能是醋氨酚中毒致肝细胞坏死的重要因素之一。 Objective: Our aim was to investigate the relationship between expression of Fas/Fas ligand(Fas L), and dose of acetaminophen, and their roles in hepatocyte apoptosis in rat . Methods: Fas/Fas L were determined in liver tissues of rats by using immunohistochemistry method. Results: Expressions of Fas/FasL in most liver tissues were both found,The positive rate was 93.3%. We found a close relationship between expressions of Fas/Fas ligand and dose of acetaminophen(P<0.05).A significant positive correlation was found between dose of acetaminophen and expression of Fas/FasL(r=0.425/0.461) . The more dose of acetaminophen was, the stronger Fas/Fas L expression was.It was significantly that some hepatocytes expressed both Fas and Fas L,and many Fas and Fas L positive cells did not accompany with liver infiltrating lymphocytes. Conclusion: Expressions of Fas Ag and Fas L on hepatocyte induced by acetaminophen may play important roles in apoptosis and death of hepatocyte . Damage of hepatocytes may be induced by autocrine or paracrine and fratricide mechanisms mediated by the hepatocytes themselves . It is one of impotant pathogenesis that may result in hepatocyte necrosis induced by acetaminophen.
