摘要
为了研究rhGM CSF和rhIL 4诱导的髓性白血病细胞刺激自体T细胞的增殖作用和IFN γ的分泌作用 ,运用rhGM CSF和rhIL 4在体外进行CML和AML外周血MNC培养 ,培养的第 7天运用流式细胞术测定CML和AML细胞的CD80 ,CD86和HLA DR表达 ;以诱导的白血病细胞作为刺激细胞 ,自体T细胞作为反应细胞 ,在体外进行混合淋巴细胞培养 (MLR) ,测定自体T细胞的增殖情况和IFN γ的分泌情况。结果显示 ,rhGM CSF和rhIL 4明显上调CML细胞HLA DR ,CD80和CD86的表达 ,并明显上调AML细胞的CD80与CD86表达 ;诱导后的白血病细胞刺激自体T细胞明显的增殖和促进IFN γ的分泌 (CML组 )作用。结论 :rhGM CSF和rhIL
To observe the proliferation of T lymphocytes stimulated by CML and AML cells which were induced by rhGM CSF and rhIL 4, and the secretion of IFN γ from proliferated T lymphocytes, the expression of CD80, CD86 and HLA DR on CML and AML cells induced by GM CSF and IL 4 was assayed by flow cytometry in vitro. Then one way mixed lymphocyte reaction was carried out, with induced leukemia cells as stimulating cells and auto T lymphocytes as reactive cells. The secretion of IFN γ from T lymphocytes was determined by double antibody sandwich ELISA. The results showed that GM CSF and IL 4 significantly upregulated the expression of CD80, CD86 and HLA DR on CML cells and CD80 and CD86 on AML cells, which could stimulate the T lymphocyte proliferation and high secretion of IFN γ (in CML group) of autologous T lymphocytes. It is concluded that the CML and AML cells induced by GM CSF and IL 4 have the ability to present tumor specific antigen to auto T lymphocyte.
