TGF-β_1、TGF-β_1RⅡ、NF-κB在肝细胞癌血管形成中的作用及机制探讨 被引量:18

Studies on the mechanisms of TGF-β_1、TGF-β_1RⅡ and NF-κB on the angiogenesis In patients with hepatocellular carcinoma

摘要 目的 探讨转化生长因子 β1(TGF - β1)、转化生长因子 β1Ⅱ型受体 (TGF - β1RⅡ )、核转录因子 -κB(NF -κB)在肝细胞癌 (HCC)血管形成中的作用及其机制。方法 用免疫组化技术 ,分别检测 36例肝癌组织和癌旁肝组织中TGF - β1、TGF - β1RⅡ及NF -κB蛋白的表达 ;以CD3 4 标记血管内皮细胞 ,观察TGF - β1及TGF- β1RⅡ蛋白与微血管密度 (MVD)的关系 ;同时观察NF -κB与TGF - β1的关系。结果  (1)肝癌组织TGF - β1蛋白表达明显高于癌旁组织 (P <0 0 1) ;肝癌组织TGF - β1RⅡ蛋白表达明显低于癌旁组织 (P <0 0 1)。(2 )肝癌组织中MVD明显高于癌旁组织 (P <0 0 1)。(3)肝癌组织中TGF - β1蛋白阳性表达与MVD呈正相关 ;TGF -β1RⅡ蛋白阳性表达与MVD呈负相关。 (4)NF -κB蛋白阳性表达与TGF - β1蛋白阳性表达呈正相关。结论 肝癌细胞中TGF - β1、TGF - β1RⅡ、NF -κB蛋白表达异常 ,并与MVD相关 ,提示它们在肝细胞癌血管形成中发挥重要作用 ,NF -κB可能在介导TGF - Objective To explore the mechanisms of TGF-β 1?TGF-β 1RⅡ and NF-κB on the angiogenesis in patients with hepatocellular carcinoma.Methods The expression of TGF-β 1 ?TGF-β RⅡ and NF-κB protein in 36 cases of HCC and surrounding HCC tissue was separately detected using immunohistochemistry technique.To observe the relationship of TGF-β 1 protein and MVD, TGF-β 1RⅡ protein and MVD, NF-κB and TGF-β 1 protein, using CD34 labelling vessel endothelial cell. Results The expression of TGF-β 1 and MVD in HCC tissue was higher than that in surrounding HCC tissue (P<0 01). But TGF-β 1RⅡprotein was on opposite way(P<0 01). There were significant positive relativity between TGF-β 1 protein and MVD, NF-κB protein and TGF-β 1 protein,but the expression of TGF-β RⅡprotein was negatively correlated with that of MVD in HCC tissue. Conclusions The expression of TGF-β 1 ?TGF-β 1RⅡand NF-κB protein in HCC tissue is abnormal. There is significant relativity between above factors and MVD. It suggests that they may play important roles in hepatocellular carcinoma angiogenesis. NF-κB may cause TGF-β 1 activation and production.
