摘要
目的 探讨膈肌纤维肌质网是否存在非序列依赖性DNA结合蛋白 ,并观察它与膈肌功能的关系。方法 Wistar大鼠随机分为对照组、急性气胸组和心痛定组 ,差速离心法分离大鼠膈肌纤维肌质网 (sarcoplasmicreticulum ,SR)膜蛋白 ,采用Western免疫印迹技术分离出SR非序列依赖性DNA结合蛋白 ,并观察此蛋白在急性气胸和给予心痛定时的变化特征。结果 大鼠膈肌SR上存在非序列依赖性DNA结合蛋白 ,相对分子质量分别为 6 0 0 0 0道尔顿和 780 0 0道尔顿。急性气胸时和给予心痛定时 ,非序列依赖性DNA结合蛋白的量有变化。结论 大鼠膈肌纤维SR上存在非序列依赖性DNA结合蛋白 ,该DNA结合蛋白可能与膈肌功能状态和Ca2 + 信号系统密切相关 ,它在膈肌和骨骼肌的病理生理过程中起重要作用。
? Objective To investigate weather the DNA binding proteins exist in the sarcoplasmic reticulum (SR) of rat diaphragm. Method The SR membrane proteins were obtained by differential centri- fugation.Western immunoblotting technique was used for detecting the presence of DNA binding proteins.The changes in the DNA binding proteins were examined in acute pneumothorax and after the administration of a Ca 2+-blocker (NIFIDINE). Result There were DNA binding proteins in the SR of rats, and the molecular weight was 78 000 and 60 000 dalton. The DNA binding proteins underwent changes in acutepneumothorax and with Ca 2+ -blocker. Conclusion The DNA binding proteins exist in rat SR and change in response to acute pneumothorax and Ca 2+ -blocker. These findings have corroboratedan activation mechanism involving the absorbing and releasing of Ca 2+ in the diaphragm and skeletal muscle SR of rats. 〔
