摘要
目的比较 3种来源的艾氏腹水癌 (Ehrlichascitescarcinoma,EAC)细胞蛋白质的特性。方法采用SDS 聚丙烯酰胺凝胶电泳 (polyacrylamidegelelectrophoresis ,PAGE)和免疫组织化学染色的方法观察 3种来源的EAC细胞蛋白质组成及Gai、TrkA、PDGF A、CDK2和NF κBp6 5抗体蛋白质表达。 结果电泳结果显示 ,北京来源的EAC蛋白质条带数最多 (2 8条 ) ,其次是山东来源的EAC(2 5条 ) ,武汉来源的EAC最少 (2 2条 ) ;免疫组织化学染色法测定 3个来源的EAC细胞的Gai、TrkA、PDGF A、CDK2和NF κBp6 5抗体蛋白质表达 ,结果不同来源的EAC与抗体的反应情况不同 ,山东来源的EAC 5种抗体的阳性表达分别为 0 %、 2 3.0 %、 0 %、 8.5 %和83.3%,北京来源的EAC分别为 77.8%、 6 .0 %、 9.0 %、 2 4 .0 %和 91.4 %,武汉来源的EAC分别为78.3%、4 .8%、8.0 %、2 7.8%和 10 0 %。结论 3种来源的EAC细胞蛋白质组成不仅有定量差异 ,而且有定性差异。
ObjectiveTo study the possible differences in some proteins among Ehrlich ascites carcinoma(EAC) strains from three sources.MethodsWith SDS polyacrylamide gel electrophoresis (SDS PAGE ) and immunohistochemical method,the protein compositions and positive expression of Gai,TrkA,PDGF A,CDK2 and NF κB p65 were observed in different EAC strains.ResultsThe protein migration bands number of EAC strain from Beijing source was the most,Shandong was next and Wuhan was least. The positive expression for Gai,TrkA,PDGF A,CDK2 and NF κB p65 were respectively 0 % , 23.0 % , 0 % , 8.5 % and 83.3 % in Shandong source,those in Beijing source were respectively 77.8 % , 6.0 % , 9.0 % , 24.0 % and 91.4 % , those in Wuhan source were respectively 78.3 % , 4.8 % , 8.0 % , 27.8 % and 100 % . ConclusionThe results indicate that some protein compositions of the EAC cells from three sources do not only have difference in quantity,but also in quality.
