摘要
目的 探索微波辐射对神经干细胞增殖、分化及凋亡的影响 ,为微波辐射的生物学效应提供基础资料。方法 利用细胞培养、流式细胞仪和细胞凋亡检测等技术 ,观察了微波辐射对神经干细胞增殖率的影响 ;5 %胎牛血清诱导微波辐射后的神经干细胞分化 ,检测分化为神经元和胶质细胞比例及细胞凋亡情况 ,并观察了细胞的形态学变化。结果 微波辐射后神经干细胞的增殖率明显下降 ,细胞凋亡数明显增加 ,分化的神经元和胶质细胞的突起变短变粗、数量减少 ,但神经元和胶质细胞的分化比例未见明显变化。结论 微波辐射对神经干细胞有较大影响 。
Objective To observe the effects of microwave radiation on proliferation, differentiation and apoptosis of human neural stem cells(NSCs), so as to provide basic data on biological effects of microwave. Methods Proliferation of NSCs was observed after microwave radiation by cell culture, flow cytometry and TUNEL method. After the differentiation of NSCs induced by microwave radiation with 5% fetal bovine serum, the ratio of the differentiated neurons to glia cells, cell apoptosis and morphological changes were detected. Results After microwave radiation, proliferation of NSCs decreased significantly, but count of cell apoptosis increased significantly. Decreased shorter and thicker neurites of neurons and glia cells were found. There were no significant changes of the ratio of the differentiated neurons to glia cells. Conclusion Power dependent microwave radiation may have great effect on neural stem cells.
