摘要
目的 观察体外培养的神经干细胞是否可以被诱导分化为类胰岛样组织。方法 体外培养神经干细胞 ,在胰岛素等外源性复合成分 (ITS)诱导分化后 ,分别进行胰腺上皮干细胞特异性标志物CK 19、胰腺 β细胞特异性成分胰岛素和胰腺α细胞特异性成分胰高糖素等的免疫细胞化学染色鉴定。结果 体外培养的神经干细胞经诱导分化 3d后 ,在神经干细胞集落附近可见散在或成群分布的上皮样细胞 ,并呈CK 19免疫阳性反应。继续培养 2~ 4周后 ,上皮样细胞增多 ,细胞圆形或呈鹅卵石样排列 ,进一步增生形成类胰岛样组织 ,其中大多数细胞呈胰岛素免疫反应阳性 ,少数细胞呈胰高糖素免疫反应阳性。结论 体外培养的神经干细胞具有向类胰岛样组织分化的潜能 ,它能分化为胰岛的α细胞和 β细胞。
Objective To observe whether the neural stem cells can be induced to differentiate into pancreatic islet like tissue in vitro . Methods The neural stem cells cultured in vitro were induced to differentiate with insulin complex components (Insulin Transferrin Selenium, ITS). The characteristics of the differentiated cells were identified by the marker of the pancreatic epithelial stem cell cytokeratin 19(CK 19), the marker of pancreatic β cell insulin, and the marker of pancreatic α cell glucagon by immunocytochemical staining. Results The neural stem cells cultured in vitro were differentiated into some epithelioid cells after having been induced by ITS for 3 days. Scattered or gathered distribution of the epithelioid cells with CK 19 immunoreactive positive was found near the colony of neural stem cells. After having been cultured for additional 2 to 4 weeks, the epithelioid cells increased to form pancreatic islet like tissue. Most of the epithelioid cells were of insulin immunoreactive positive, but only a few were of glucagons positive. Conclusion The neural stem cells cultured in vitro are potent to differentiate into pancreatic islet like tissue and they can differentiate into pancreatic β and α cells.
