摘要
目的 :改变手工试管赖氏法操作使之成为自动化条码传送微板赖氏法的操作 ,同时为了克服轻度脂血对结果的影响。方法 :采用方阵法和正交试验设计方法 ,筛选微板赖氏法最佳浓度和加液量 ;利用双波长等吸收的特点 ,消除轻度脂血吸收的干扰。结果 :液体总量有赖氏试管法的 610 0 μl缩小为赖氏微板法的 2 70 μl;采用双波长 492nm、63 0nm检测 ;该法检测 2 0 0 1年卫生部临检中心发放的ALT室间质控物品 ,准确度为 10 0 %。结论 :本研究填补了国内血站系统赖氏法ALT长期手工操作的局面。该法省工省时 ,操作简便 。
