摘要
目的 :检测脑胶质瘤细胞体外药敏及P -糖蛋白的表达 ,为临床选择化疗药物制定方案提供依据。方法 :采用MTT法检测 30例脑胶质瘤患者肿瘤细胞 ,对化疗药物的敏感性以及采用免疫组化方法检测脑胶质瘤细胞的P -糖蛋白的表达。结果 :30例脑胶质瘤患者肿瘤细胞体外药敏显示 :高三尖杉酯碱 (H)、长春新碱(VCR)、阿糖胞苷 (Ara -C)、足叶乙甙 (Vp - 16 )、柔红霉素 (DNR)、米托葸昆 (MX)、平阳霉素 (SP)、阿克拉霉素 (ACR)、阿霉素 (ADM )、环已亚硝脲 (CCNU)等化疗药物对肿瘤细胞均有一定的抑制作用 ,以CCNU、ADM、ACR、DNR、MX、Vp - 16抑制作用较强。检测 2 8例患者脑胶质瘤细胞P -糖蛋白表达 ,2 4例表达(- ) ,4例表达 (+) ,其中 3例为复发患者。结论 :胶质瘤细胞体外药敏及P -糖蛋白表达的检测 ,可为临床指导用药 ,制定化疗方案 ,提供重要参考实用价值。
Objective:To detect the chemosensitivity test and p-gp expression of glioma cells, to give evidence to select anti-tumor drug and design chemotherapy programme. Methods: Drug sensitivity of tumor cells in 30 patients with glioma was detected by MTT assay, and multidrug resistance p-gp was tested by immunohistochemical assay using a monoclonal antibody.Results: The results showed that H, VCR, Ara-c, Vp-16, DNR, MX, SP, ACR, ADM? CCNU had inhibitory effect to tumor cells, especially CCNU, ADM, ACR, DNR, MXa and Vp-16. In 28 brain glioma cases, 24 cases were negative and 4 cases were positive, 3 of 4 cases relapsed. Conclusions:It is important that the chemosensitivity test of glioma and the expression of p-gp should be used to help us to select anti-tumor drug and design chemotherapy protocols.
