摘要
目的 :研究白细胞介素 - 2 (IL - 2 )和白细胞介素 - 12 (IL - 12 )活化骨髓的抗肿瘤效应与重建造血功能 ;探讨活化骨髓的性质和杀伤肿瘤细胞的机制。方法 :测定活化骨髓杀伤肿瘤细胞的活性 ;检测活化骨髓的细胞表面分化抗原 ,特别是CD34+/CD38-细胞群的表达。结果 :活化骨髓与L12 10 细胞、新鲜白血病细胞共同培养后显示有杀伤肿瘤细胞作用 ,以培养 3~ 4天时最强 ;活化骨髓细胞的CD3、CD8、CD16、CD5 6分化抗原表达与新鲜骨髓相比升高 ,有显著性差异 (P <0 0 1)。L12 10 细胞、新鲜白血病细胞与活化骨髓共同培养后分化抗原CD71、CD95表达下降 ,超二倍体、亚二倍体等异倍体减少 ,与对照组相比有显著性差异 (P <0 0 5 ) ;活化骨髓中CD34+/CD38-细胞分化抗原表达与新鲜骨髓相比没有显著性差异 (P >0 0 5 )。IL - 12活化骨髓效靶比 (E∶T)稀释一倍后杀伤白血病细胞活性仍然比IL - 2活化骨髓明显 ;IL - 2联合IL - 12诱导后活化骨髓抗肿瘤活性进一步增强。结论 :活化骨髓具有抗肿瘤效应 ,其中有细胞毒T细胞、自然杀伤细胞的作用 ,也有凋亡机制参与 ;活化骨髓还保留有重建造血的功能 ;IL - 12活化骨髓比IL - 2活化骨髓的活性高 ;且IL - 2联合IL -
Objective:To study Interlukin 2(IL 2)and IL 12 activated bone marrow anti tumor effect and the function of hempopoietic reconstruction; To study the nature of activated bone marrow and mechanisms of kill tumor cell.Methods:Assay the competence of activated bone marrow kill tumor cell; Detect cell surface differentiation antigen of activated bone marrow, particularly the expression of CD34+/CD38 cell group.Results:After activated bone marrow and L 1210 cell, fresh leukemia cell being cultured together, Action for kill tumor cell display, while 3 4 day the highest; Compared differentiation antigen expression of activated bone marrow CD3, CD8, CD16, CD56 with fresh bone marrow, difference is significant ( P <0 01). After L 1210 cell,fresh leukemia cell and activated bone marrow, being cultured together, differentiation antigen CD71, CD95 expression decreases, heteroploid (hyperdiploid,hypodiploid) reduces, there is significance in the difference compared with control group ( P< 0 05).Activated bone marrow CD34+/CD38 cell group differentiation antigen expression compares with fresh bone marrow, no significant difference ( P >0.05). After IL 12 activated bone marrow effect target ratio (E∶T)dilution 1 time, the action of kill leukemia cell still higher than IL 2 Activated bone marrow.Conclusion:There are anti tumor effects in the activated bone marrow, among cytotoxic T cell effect, nature killer cells action with the apoptosis mechanism together; Activated bone marrow retains the function of hemopoietic reconstruction. IL 12 activated bone marrow effects are higher than IL 2 activated bone marrow and was further increased when IL 2 in combination with IL 12.
