血清中游离脂肪酸的RP-HPLC分析

摘要 用乙醇除去血清中与游离脂肪酸(FFA)结合的蛋白,在1 EDC·HCl的催化下,以2 NPH·HCl为衍生剂,用正己烷萃取,然后采用改进的反相高压液相色谱法(RP HPLC)进行色谱分离,结果表明,沉淀蛋白的最佳条件是乙醇为14.3mol L,温度为60℃,流动相为乙腈 水,其浓度为4.88mol L,柱温为30℃,流速为1.2mL min,紫外吸收波长为406nm,pH=4.5.实验分析结果表明,正常人之间的血清FFA水平基本一致,二型糖尿病(T2DM)患者的血清FFA水平显著增高(P<0.01). Eight free fatty acids (FFA) in serum removed protein were separated by RPHPLC method, and the conditions were: 2NPH·HCl as derivative, 1EDC·HCl as catalyst,and the hexane for extraction.The results showed that the optimum conditions for precipitating the protein were that the concentration of alchohol was 14.3 mol/L,the temparature was 60?℃; ratio of ethylnitrile and water was 4.88 mol/L,column temparature was 30?℃,current velocity was 1.2?mL/min, ultraviolet absorbent wavelength was 406 nm,pH=4.5 for derivation.The concentration of FFA in serum among healthy persons was basicly agreement, while that among T2DM patients had a clear increment(P<0.01).
