自体半脾组织移植的实验研究

摘要 将145只小鼠随机分组,Ⅰ对照组(25只),Ⅱ半脾保留组(40只),Ⅲ脾移植组(55只)和Ⅳ全脾切除组(25只)。光镜、TEM和SEM检查显示移植脾成活。显微分光光度计测单位面积酶 光值,ACP值Ⅰ组(0.462±0.013),Ⅱ组(0.304±0.009)和Ⅲ组(0.180±0.014)顺次显著降低(P<0.05);ANAE值Ⅰ组(0.260±0.021)大于Ⅱ组(0.218±0.016)和Ⅲ组(0.210±0.022) (P<0.05),后两组无显著差异(P>0.05)。微量全血恒温法测化学发光值,有脾鼠血清调理者Ⅰ、Ⅱ、Ⅲ组逐渐降低(P<0.05),Ⅲ、Ⅳ组无差异(P>0.05);无脾鼠血清调理者Ⅰ组大于Ⅱ、Ⅲ、Ⅳ组,Ⅳ组小于Ⅱ、Ⅲ组(P<0.05)。结果提示,移植脾有一定的免疫吞噬功能。 One hundred and forty-five mice were randomized into 4 groups:Group Ⅰ consisted of 25 mice with sham operation,group Ⅰ of 40 mice with hemisplenectomy,group Ⅲ of 55 mice with hemisplenic autotransplantation,and group Ⅳ of 25 mice with splenectomy.Twelve weeks after operation,it was found under optical microscopy,transmission electron microscopy and scanning electron microscopy that the autografted hemispleen was viable.The contents of alpha-naphthol acetylate esterase (ANAE) and acid phosphatase (ACP) in the splenic tissue were measured with microspectrophotometer.ACP level decreased continuously in group Ⅰ (0.462±0.013),group Ⅱ (0.304±0.009),and group Ⅲ(0.180±0.014) (P<0.05).ANAE level was lower in group Ⅲ(0.210±0.022) than in group Ⅰ (0.260±0.021) (P<0.05) and was similar between group Ⅲ and group Ⅰ (0.218±0.016).The data of chemiluminescence(CL)assay opsonized with normal mice serum dropped significantly in groupsⅠ,Ⅱ,Ⅲ and Ⅳ,while the data of CL assay opsonized with asplenic miceserum was significantly higher in group Ⅲ than in group Ⅳ,and significantlylower in group Ⅲ than in group Ⅰ (P<0.05),and similar between group Ⅲand group Ⅱ.The findings suggest that the autografted hemispleen possesses certain capability of phagocytosis and immune functions.
