美金刚(Memantine)对敌敌畏中毒小鼠的实验治疗研究 被引量:9

摘要 目的研究非竞争性N -甲基 -D -天门冬氨酸 (NMDA)受体拮抗剂美金刚对有机磷农药敌敌畏中毒小鼠的治疗作用。方法根据正交实验设计原理 ,选用L9(34)正交表 ,以肌束震颤出现时间、翻正反射消失时间和存活时间为指标 ,分别测试美金刚和/或阿托品对敌敌畏染毒小鼠的治疗效果。结果美金刚可明显延长染毒小鼠的存活时间、肌束震颤出现时间 ,并可部分改善肌束震颤症状 ;与阿托品联合使用后效果更加明显 ,提示两药及早联合用药时治疗效果更好。结论美金刚和阿托品联合使用对敌敌畏中毒有很好的协同治疗作用 ,可明显改善肌束震颤症状。鉴于美金刚与阿托品作用机制不同 ,建议可在临床上试用。但在中毒指征及给药剂量的正确掌握方面 。 Objective The purpose of this study was to check whether Memantine could serve as an antagonist against the acute toxicity of Dichlorvos in male Kun-ming mice. Methods The therapeutic schedules of Memantine and atropine were arranged according to the L9(34) orthogonal test table. The survival time, the loss of righting reflex and the latency of onset of muscle fasciculations were observed. Results A single dose of Memantine could prolong the survival time and the latency of onset of muscle fasciculations, meanwhile it could also reduce the intensity of muscle fasciculations. The much better results were observed when it is given combined with atropine. Conclusion The results showed that, the combined treatment of Memantine and atropine could effectively antagonize the acute toxicity of Dichlorvos, and improve the intoxicated signs such as the intensity of muscle fasciculations. These indicated that the use of Memantine in the clinical treatment of organphosphates intoxication sounds reasonable and acceptable. Using cautiously, therapeutic effects of Memantine combined with atropine would be expected in the treatment of organophosphates intoxication.
