复方丹参方对拟血管性痴呆大鼠脑内神经递质的影响

Effect of Compound Danshen(复方丹参方) on Transmitter in Rat Brain of Vascular Dementia

目的 :探讨复方丹参方对拟血管性痴呆大鼠的治疗作用机理。方法 :Wistar雄性大鼠经夹闭双侧颈总动脉 (CCA) ,并腹腔注射硝普钠 (2 .5 mg· kg- 1 )造模。选术后 7d存活的有显著学习记忆障碍的大鼠 4 0只为血管性痴呆模型 ,随机分为 4组。西药组给予喜得镇悬浮液 (0 .5 mg· kg- 1 · d- 1 ) ,复方丹参大小剂量组给予复方丹参方稀释液 (剂量分别为 10 g· kg- 1 ·d- 1、5 g· kg- 1· d- 1 ) ,模型组与正常组均给予等容量生理盐水灌胃。连续 2 1天。结果 :模型组与正常组比较 ,模型组大鼠脑内 Ach、5 - HT含量明显降低 ,Ach E活性明显升高 (P<0 .0 1)。复方丹参方大剂量组能明显升高 Ach含量及 5 - HT含量 ,降低脑内 Ach E活性 (P<0 .0 5 ,0 .0 1) ;复方丹参方小剂量组亦有类似作用 ,但 P>0 .0 5。与西药组比较无显著差异 (P>0 .0 5 )。结论 :复方丹参方大小剂量均能改善胆碱能系统的功能 ,调节脑组织 Ach生理代谢 。

Objective:To explore the mechanism of treatment of Compound Danshen Formula(CDF,复方丹参方) on vascular dementia.Method:Forty rats modeled as vascular dementia were selected and randomly divided into four gruops.Western drug group(Group A) were treated with Hydergine(0.5 mg·kg -1 ·d -1 );CDF group (Group B) were treated with CDF diluent (respectively 10 g·kg -1 ·d -1 ,5 g·kg -1 ·d -1 );The model group (Group C)and the normal group (Group D) were treated with saline.Results:After 21 days,the content of Ach,5 HT in rat brain in model group decreased and the activity of AchE increased(P<0 01).High dosage of CDF could markedly increase the content of Ach and 5 HT,and decrease the activity of AchE (P<0 01).Low dosage of CDF had the similar effect but no significance (P>0 05).Conclusion:Both high and low dosage of CDF had the effect of improving the function of cholinergic nerve system in brain and regulating the metabolism of Ach to improve memory function.
