摘要
为探究先天性心脏病(先心病)伴肺动脉高压(肺高压,PH)患儿肺组织一氧化氮合酶(eNOS)的表达有否改变,随机选择法洛四联症(tetralogy of Fallot,TOF)、单纯室间隔缺损 (ventricular septal defect,VSD)/房间隔缺损(atrial septal de-fect,ASD)、先心病合并PH患儿48例,分成3组,取少许右肺中叶组织,利用免疫组织化学法对eNOS进行半定量分析。结果显示先心病伴PH组患儿肺组织内皮细胞内的eNOS染色明显弱于TOF组和单纯VSD/ASD组患儿(P<0.01)。提示先心病PH组肺组织的eNOS含量水平低下,造成内源性NO生成减少,为临床吸入NO治疗PH提供了理论依据。
To explore the synthesis change of endothelial nitric oxide synthase (eNOS) in lung tissue in children with congenital heart disease (CHD) complicated pulmonary hypertension (PH), eNOS in lungs was determined by semiquanti-lalive immunohistochemical method in 48 CHD children who were randomly divided CHD with PH group as group I , Tetralogy of Fallot group as grup 0 and VSD/ASD group as grooup III respectively. The results showed that eNOS synthesis in group I was significantly lower than those in other two control groups (P<0.01) . It is suggested that these is a lower eNOS and NO levels in pulmonary vascular endothelial cell in CHD children with PH. It will be feasible to treat with inhaled NO that can provide a new widely applicable prophylactic approach for the prevention of postoperative PH in CHD children.
