摘要
作者利用每3 d换新鲜培养液并重新加相应浓度药物的长期液体培养方法,研究了低浓度(1.88×10^(-5)mmol/L)三尖杉酯碱(Harr)对人早幼粒白血病细胞(HL - 60)系的诱导分化作用,结果表明低浓度(1.88×10^(-5)mol/L)Harr与HL -60细胞作用12 d,细胞形态,NBT还原试验,酵母多糖微粒吞噬及细胞超微结构出现明显成熟分化,且很少有细胞毒作用。为进一步研究Harr对HL -60细胞的诱导分化机理提供了一种理想的方法。
Low concentration of Harringtonine (Harr, 1.88 × 10 -5 mmol/L )on induction of differentiation and growth inhibition of the promyelocytic leukemia cells (HL - 60 )by the method of long- term culture (modified by fresh medium which contained the same concentration of drug as initiation of culture at 3 day intervals during the culture )was studied .The results showed that the HL - 60 cells by incubation in low concentration of Harr (1-88 × 10 -5 mmol/L ) for 12 days were induced to differentiate into monocyte/macrophage cells morphologically and functionally when the proliferation of leukemia cells was still more than 90% as assessed by typan blue dye exclusion test . This result may be useful for further studying of the effect of low concentration of Harr on induction of differentiation of HL - 60 cells .
