更昔洛韦对转染TK基因的人骨髓瘤细胞的生长抑制作用及旁观者效应

摘要 目的 :探讨更昔洛韦对转染 TK基因的骨髓瘤细胞生长的抑制作用。方法 :应用缺陷型反转录病毒介导的 TK基因系统 (RV- HSV- TK)转染骨髓瘤细胞 ,采用细胞增生实验 (MTT法 ) ,研究更昔洛韦对转染 TK基因的骨髓瘤细胞株的抑制作用。结果 :转染了 TK基因的骨髓瘤细胞的生长曲线与未转染 TK基因的细胞无差异。 0 .1μg/ m L~ 5 0 μg/ m L 的更昔洛韦对转染 TK基因的骨髓瘤细胞有明显的抑制作用 ,且与更昔洛韦的作用时间成正比。将转染阳性细胞与未转染细胞共孵育 ,加入 10 μg/ m L 的更昔洛韦作用 96 h,可见旁观者效应。结论 :采用 RV- HSV- TK系统转染骨髓瘤细胞 ,有较高的转染率 ,转染了 Objective:To illustrate the effect on myeloma cells with TK gene by GCV.Methods:By using RV HSV TK gene system and MTT technique, the growth inhibited effect on myeloma cells with TK gene by GCV were analysed.Results:TK gene was successfully transmitted into Myeloma cell lines by retrovirus. GCV showed significant toxicity on myeloma cells with TK gene which were dose concentration(between 0.1 μg/mL to 50 μg/mL) and time dependent. We also observed by stander effect after coincubation of myeloma cells with it's TK gene transfected cells.Conclusion:It was very efficient for myeloma cells to be transferred TK gene with retrovirus and these cells were very sensitivity to GCV.
