摘要
研究拉伸应变和加载时间对体外培养人牙周膜成纤维细胞 ( PDL F)的白细胞介素 - 1β( IL - 1β)分泌量的影响。将原代、传代培养成功的 PDL F制备成实验用细胞 ,分为 5组 ( n=3 ) ,而后分别置于特制的加力装置上拉伸加载 ,加力至拉伸应变量分别为 0 %、8%、12 %、16%和 2 0 % ,加力时间为 2 4、48、72 h。用双抗 EL ISA法分析 IL - 1β的含量。结果表明 :8%、12 %和 16%拉伸应变组 IL - 1β的分泌量随着加力时间和力值的增加而递增 ;在各个时间组 ,IL - 1β分泌量在 16%应变值处达到最高值 ;2 0 %拉伸应变组 ,加力 2 4h和 48h,IL - 1β分泌量明显高于 0 %应变组 ,与 12 %和 16%应变组相比已开始下降 ,加力 72 h,IL - 1β分泌量大幅度下降 ,已低于对应的 0 %应变组。在细胞生理应变承受范围内 ,静态机械拉伸应变能促进人牙周膜成纤维细胞 IL - 1β的分泌 。
This study aims at exploring the effects of tensile strain and loading time on the secretion of IL-1β of human periodontal ligament fibroblast. Five tensile strain values including 0%?8%?12%?16%?20% and three loading time including 24h, 48h, 72h are set in this study. The prepared cell samples are mounted on the self-devised loading apparatus in vitro. The content of IL-1β in each sample was determined using double-antibody ELISA. The secretion amount of IL-1β per day is directly proportional to the loading time and tensile strain value in tensile strain group of 8%?12%?16%.The secretion amount of IL-1β reaches its maximum at tensile strain value of 16% in loading time groups. Loaded strain for 24h and 48h, the secretion amount of IL-1β at tensile strain value of 20% is obviously more than that at value of 0%, but the amount already begin to decrease apparently. Loaded strain for 72h, the secretion amount of IL-1β decreases to a great extent that it is less than the amount at strain value of 0%. The tensile strain stimulates the secretion of IL-1β by human periodontal ligament fibroblast when the strain is under normal physiological extent, but the stimulation effect fades out as time goes on.
