摘要 作者用培养的原代心肌细胞,建立缺氧再灌注模型.对缺氧60,120,180 min及缺氧120 min后再给氧30,60 min时,心肌细胞培养液中的ALP,CPK,GOT,GPT诸酶含量的变化及降钙素基因相关肽(CGRP)对这一过程的影响进行了观察.结果显示:细胞内ALP,CPK,GOT,GPT的释放量随缺氧及再灌注时间的延长逐渐升高.缺氧早期ALP,CPK反映细胞损伤的程度较GOT,GPT敏感.10^(-9)和10^(-8)mol/L CGRP可明显减少缺氧120 min时酶的漏出,而10^(-7)mol/L则无此作用,10^(-8)mol/L CGRP可明显减少心肌细胞缺氧再灌注时上述酶的漏出,说明CGRP可减轻心肌细胞缺氧灌注时的损伤,对心肌细胞具有保护作用. In the present experiment, an anoxic-reperfused animal model was established by using cultured primordial cells and the ALP, CPK, GOT and GPT content in the myocardial culture was measured 60, 120, and 180 min after anoxia and 30 and 60 min after reperftission and the effects of calcitonin gene related peptide (CGRP)on the anoxic-reperfused myocardial cells were studied. The findings indicated that the release of ALP, CPK, GOT and GPT from the cells gradually increased as anoxia and reperftission prolonged. In the early period of anoxia, the level of ALP and CPK was a more sensitive indicator of the extent of injury to the myocardial cells than that of GOT and GPT. CGRP at concentrations of 10-9 and 10-8 mol/L was able to significantly reduce the leakage of enzymes 120 min after anoxia while CGRP at a concentration of 10-7 was not. The finding that CGRP at appropriate concentrations might reduce the leakage of enzymes demonstrated that CGRP might protect against damages to anoxic-reperfused myocardial cells.
