摘要
将人脐带血管内皮细胞(HEC)培养于人羊膜基膜模型上,构成一种新的血管内皮模型,并与利用人胎儿主动脉内膜等多种血管内皮模型作对比观察。从粘附率,浸润力及形态学角度比较研究了大肠癌细胞及其多细胞球体对血管内皮细胞的粘附和浸润作用。结果表明:癌细胞直接浸润血管内皮细胞是一种重要的浸润血管方式。多细胞球体对血管内皮具有高粘附,高侵袭特性。
Seeding onto the basement membrane ( BM ) of a HABM ( Human amniotic basement membrane ) model, the human endothelial cells derived from the umbilical vein ( HEC ) can form confluent monolayers on the human BM surfaces in the presence of ECGS. Using the new vascular endothelium model, the adhesion, invasi-veness and other biological characteristics of the colorectal cancer cell lines ( HR-8348, HCe-8693 ) and HR-8348 spheroids to the HEC were studied comparatively. The results showed that the carcinoma cells can adhere and invade the HEC directly, even completely penetrating it. The invasiveness and adhesion of the spheroids was significantly greater (P<0.01)than that of the corresponding single cell suspension. These findings suggested that the HEC may play animportant role in facilitating invasion, and the spheroids may be the major elements in the process of metastasis.
