摘要
以黄花梨为试材,建立了梨树RAPD分析的优化反应条件,即25μL反应体系中含有Tris-HCl(pH9.0)10mmol/L,KCl50mmol/L,MgCl22.0mmol/L,dNTP200μmol/L,模板DNA10ng,引物400μmol/L,TaqDNA聚合酶1.5U.适宜的扩增程序为94℃120s,1个循环;94℃60s、37℃75s、72℃120s,45个循环;72℃10min,1个循环.
The Huanghuali(Pyrus pyrifolia Nakai) was used to establish to optimize RAPD reaction condition of the pear.The total volume (25 μL) were contained 10 mmol/L Tris-HCl(pH9.0), 50 mmol/L KCl, 2.0 mmol/L MgCl2, 200 μmol/L dNTP, 10 ng template DNA, 400 μmol/L primer and 1.5 U Taq DNA polymerase. The optimal amplification procedure was 94 ℃120 s; 1cycle:94 ℃60 s, 37 ℃75 s, 72 ℃120 s, 45 cycles; 72 ℃10 min.
