摘要
采用随机扩增多态性DNA(RAPD)标记,对黑麦属(SecaleL)7个种共12份材料进行了遗传多样性检测。结果表明,被测材料间RAPD标记多态性较高。40个随机引物中,有25个引物(占62 5%)的扩增产物具有多态性。25个引物共扩增出167条带,其中89条带(占53 2%)具有多态性,每个引物可扩增出1~10条多态性带,平均3 6条。RAPD标记遗传距离(GD)变异范围为0 1382~0 4512,平均值为0 2712。聚类分析表明,在GD值0 3850水平上,12份材料可聚3类,S.africamum和S.sil vestre与其它材料间的遗传分化较大,分别单独聚为一类,其余10份材料则聚为一类。据此认为,RAPD标记可以作为黑麦属物种鉴定的指纹图谱。
Genetic diversity of 12 accessions of Secale L., distributed to 7 species,was analyzed based on random amplified polymorphic DNA (RAPD) markers.Fourty arbitrary primers (10mer) were used for the PCR amplification of genomic DNA, among which 62.5?% primers could produce polymorphic bands.A total of 167 bands were amplified from 25 primers,while 89 bands (about 53.2?%) were polymorphic. One to 10 polymorphic bands could be amplified from each primer,with an average of 3.6 bands.The mean genetic diversity(GD) value based on RAPD data among 12 accessions of Secale L.was 0.2712, ranging from 0.1382 to 0.4512.At the level of GD 0.385,cluster analysis showed all materials could be classified into 3 subclusters,in which S.africanum and S.silvestre were less related with others. These results suggested that RAPD markers could be used as one fingerprint in Secale.
