利用末端修补接头连接法确定PRPSAP1基因的结构(英文)

摘要 为确定磷酸核糖焦磷酸合成酶(PRS)复合物中PAP39亚单位的编码基因PRPSAP1的结构,采用了末端修补接头连接法.结果表明PRPSAP1基因由10个外显子组成,其大小分别为83bp、53bp、67bp、173bp、116bp、56bp、146bp、71bp、147bp和156bp.PRPSAP1基因结构的确定有助于进一步研究人PRPSAP1基因的功能. To determine the genomic structure of human PRPSAP1 gene encoding the PAP39 subunit of phosphoribosylpyrophosphate synthetase(PRS) complex,the End Trimming and Cassette Ligation(ETCL)PCR method was applied.The result showed that PRPSAP1 gene consists of 10 exons and the sizes of exons are 83?bp,53?bp,67?bp,173?bp,116?bp,56?bp,146?bp,71?bp,147?bp and 156?bp,respectively.Determination of PRPSAP1 genomic structure is very important for further exploring the functions of human PRPSAP1 gene.
