摘要
目的 :探讨心力衰竭患者血浆血小板活化因子 (PAF)、血小板活化因子乙酰水解酶 (PAF AH)活性的变化 ,以及外周血中单个核细胞 (PBMC)PAF受体 (PAF R)的表达。方法 :对 30例心力衰竭患者采用生物法检测血浆中PAF含量 ,酶水解底物显色法测定PAF AH活性 ,流式细胞仪检测PBMC上PAF R的表达。并与 30例健康体检者对照。结果 :心力衰竭患者血浆中PAF含量 (11.12± 1.80 ) μg L ,PAF AH活性 (2 4 .5 0± 7.86 ) μmol (min·L)、PBMC上PAF R的表达与健康体检者比较差异有非常显著性意义 (P <0 .0 1)。结论 :PAF、PAF AH、PAF
Objective:To study the concentration of platelet activating factor (PAF) and the activation of PAF Acetylhydrolase (PAF AH) in patients with heart failure as well as the expression of PAF Receptor (PAF R ) in peripheral blood PBMC of these patients. Methods: Thirty patients with heart failure and 30 healthy people were enrolled in this study. The levels of PAF were determined by the washed platelet aggregation test (bio assay) and the activation of PAF AH was determined by the changes of substrate assay. The expression of PAF R on PBMC was examined by cytoflowmetry.Results: The results showed that the concentration of PAF in heart failure group was significantly higher than that in healthy group [( 11.12 ± 1.80 )μg/L vs ( 7.86 ± 1.41 )μg/L, P< 0.01 ], whereas the activation of PAF AH in heart failure group was significantly lower than that in healthy group[( 24.50 ± 7.86 ) μmol/min·L -1 vs ( 32.14 ± 12.85 ) μmol/min·L -1 ,P< 0.01 ]. Furthermore, the expression of PAF R on PBMC increased markedly in peripheral blood of patients with heart failure (P< 0.01 ).Conclusion: The changes of PAF and PAF AH and the expression of PAF R on PBMC are related to the pathogenesis and development of heart failure. Drug of anti PAF or PAF receptor antagonists may be used in heart failure.
