摘要
目的 观察芪归合剂对地塞米松治疗的肾病综合征大鼠(NS鼠)生长障碍的作用及其可能的机制。方法 制备大鼠阿霉素NS模型,设正常对照组、NS模型组、芪归治疗组、激素治疗组和激素芪归治疗组。测量各组鼠的身长、体重、尿蛋白和血白蛋白水平。以放射免疫法和免疫放射法测各组血、尿胰岛素样生长因子(IGF)-1和IGF结合蛋白(BP)-3水平;用RT-PCR法检测肝IGF-1mRNA、IGFBP-3mRNA表达水平。结果NS模型组体重增长、身长增长、血IGF-1.IGFBP-3低于正常组,尿IGF-1高于正常组(P<0.01或P<0.05);肝IGF-1mRNA表达与正常组无差异,而IGFBP-3 mPNA表达低于正常组(P<0.01)。激素治疗组的体重增长、身长增长、血IGF-1和IGFBP-3水平、肝IGF-1mRNA、IGFBP-3mRNA表达水平低于模型组(P<0.01或P<0.05)。芪归治疗组的身长增长、体重增长、血清IGF-1、IGFBP-3水平、肝IGF-1 mRNA、IGFBP-3 mRNA表达水平高于模型组(P<0.01或P<0.05)。激素芪归组的身长增长、体重增长、血清IGF-1、IGFBP-3水平、肝IGF-1mRNA、IGFBP-3 mRNA表达水平高于激素治疗组(P<0.01或P<0.05)。结论 芪归合剂可能通过增加地塞米松治疗的NS鼠肝IGF-1 mRNA、IGFBP-3 mRNA的表达水平而部分拮抗地塞米松对NS鼠生长的抑制作用。
Objective To study the effect of Astragalus and Angelica mixture on growth failure induced by dexaraethasone in rats with nephrotic syndrome (NS) . Methods Forty six-week old male Sprague-Dawley rats with a starting weight of 160 ~ 180 g were randomly divided into five groups: control group, NS model group, NS group treated with Astragalus and Angelica mixture, NS group treated with dexamethasone, and NS group treated with Astragalus and Angelica mixture and dexamethasone. Weight, length, urine protein, and serum albuine were measured. Serum and urine concentrations of IGF-1 and IGFBP-3 were assayed by RIA and IRMA respectively; and IGF-1 mRNA and IGFBP-3 mRNA levels in liver were assayed by RT-PCR. Results Weight gain and length gain, serum IGF-1 and IGFBP-3 levels in NS model group were significantly lower than that of the control group and urine IGF-1 levels were higher than that of the control group (P < 0. 01 or P < 0. 05) . Liver IGF-1 mRNA levels in NS model group were similar to control rats, but hepatic IGFBP-3mRNA levels in NS group were reduced. Weight gain and length gain, serum IGF-1 and IGFBP-3 levels, liver IGF-1 mRNA and IGFBP-3 mRNA levels in NS group treated with dexamethsone were lower than those in NS model group( P < 0. 01 or P < 0. 05) . Weight gain and length gain, serum IGF-1 and IGFBP-3 levels, liver IGF-1 mRNA and IGFBP-3 mRNA levels in NS group treated with Astragalus and Angelica mixture were higher than those in NS model group (P < 0. 01 or P < 0. 05) . Weight gain and length gain, serum IGF-1 and IGFBP-3 levels, liver IGF-1 mRNA and IGFBP-3 mRNA levels in NS group treated with Astragalus and Angelica mixture and dexamethsone were higher than those in NS group treated with dexamethasone( P < 0. 01 or P < 0.05) . Conclusion Astragalus and Angelica mixture can partly antagonize the growth inhibition effect induced by dexamethsone in rats with nephrotic syndrome through increasing hepatic IGF-1 mRNA and IGFBP-3 mRNA levels .
