摘要
目的 探讨雌、雄激素对大鼠卵巢颗粒细胞凋亡基因表达的作用。 方法 体外培养 SD大鼠卵巢颗粒细胞设加睾酮 1μg· ml- 1 的雄激素组 ,加雌二醇 1μg· ml- 1 的雌激素组和不加性激素的对照组 ,以 MRNA差异显示技术研究各组凋亡相关基因的表达。所获差异片段以NORTHERN杂交鉴定 ,阳性片段以自动测序仪测序并与 GENBANK数据库资料比较。结果 发现 7个差异条带 ,其中两个分别与 BCL-2和BAX基因同源 ,另 5个条带为假阳性。结论 BCL-2和 BAX基因直接参与了甾体性激素作用下的大鼠卵泡颗粒细胞凋亡 ,雄激素组 BCL-2表达下调 ,BAX表达上调 ;雌激素组则相反。
OBJECTIVE To study the effects of androgen and estrogen on the gene expression related to rat ovarian granulosa cell apoptosis METHODS The differential gene expression related to the rat ovarian granulosa cells after androgen(1μg·ml -1 ), estrogen(1μg·ml -1 ) treatments were analysed by MRNA differential display, the obtained differential display fragments were further identified by nothern blot, the positive fragments were sequenced and did homology searching in Genbank RESULTS Among the obtained seven differential display fragments, two have high homology with bcl-2 and bax for each other And the other five fragments were found to be pseudo-positive CONCLUSION Both bcl-2 and bax participate straightly in the apoptosis of rat ovarian granulosa cells which affected by sex steroids Adrogen reduced the expression of bcl-2,as well as enhanced that of bax, the effect of estrogen to the two gene is inverse
