酵母突变株的筛选

摘要 目的:建立快速、简便的筛选酵母突变株的方法。方法:利用同源重组技术,通过转座子上的LacZ报告基因,从酵母基因 组文库中,将发生突变的菌株进行检测,从而筛选出酵母突变株。结果:在x-gal培养基上出现蓝色菌落,即为所需的阳性菌落。 结论:利用同源重组技术,初步筛选出所需的酵母突变株,为新的功能性基因组的研究和发展,为基因药物的研究与开发构建平台。 Objective: To establish a fast, simple method to screen mutant yeast strains. Method: Homologous recombination technology was used to detect mutant yeast strains in yeast genomic library, with reporter gene LacZ in transposon. Result: Blue strains on the X - gal medium were positive one. Conclusion: A platform used to study new functional genome is constructed by homologous recombination technology. The platformmay provide an alternative way in the development of gene medicine.
