Identification of Omeprazole Urinary Metabolites in Human by High Performance Liquid Chromatography-Electrospray Ion Trap Mass Spectrometry

摘要 应用高效液相色谱 电喷雾离子阱质谱法鉴定了人口服奥美拉唑 (OPZ)后 0~ 6h内尿中的代谢物。尿中代谢物经富集后 ,应用高效液相色谱法分离 ,然后在线进行选择离子扫描 (SIM)、二级碎片离子全扫描(fullscanMS2 )和三级碎片离子全扫描 (fullscanMS3) ,进而对尿中微量的烷烃羟基化、O 脱烷烃化、硫氧化和还原以及葡萄糖醛酸化和硫酸化的 Nine urinary metabolites of omeprazole (OPZ) in healthy subjects (0 similar to 6 hours urine samples after administration of 40 mg Losec capsule). were identified by high performance liquid chromatography-electrospray ion trap-mass spectrometry (HPLC-ESI/IT-MS(n)). The mixed urine was simply enriched by C(18) SPE column, OPZ and its metabolites were completely separated by HPLC and detected in selected ion monitoring (SIM) mode by ESI/IT-MS(n). Product ion spectra of molecular ions were recorded after collision-induced dissociation (CID) by ion trap OTMS') in full scan MS(2) and full scan MS(3) mode. Therefore, nine trace urinary OPZ metabolites (biotransformed by aliphatic hydroxylation, O-dealkylation, S-oxidation and reduction, glucuronidation or arylsulfatation) were identified by interpreting mass spectrometric data.
