摘要
目的探讨茶多酚、芦荟甙对中波紫外线(UVB)辐射引起皮肤损伤的保护机理。方法酶联免疫吸附试验(ELISA)检测UVB辐射角质形成细胞(HaCaT)引起TNF-α及IL-1β的分泌量的变化;RT-PCR法检测TNF-α及IL-1β的mRNA表达。结果UVB辐射后,HaCaT细胞分泌TNF-α及IL-1β的量比正常对照组明显增多;而应用茶多酚、芦荟甙后,分泌量则均明显降低。TNF-α及IL-1β的mRNA表达在UVB照射后明显增加;应用茶多酚、芦荟甙处理后,表达明显降低。差异均有显著性(P<0.01)。结论茶多酚、芦荟甙可以通过降低UVB引起的TNF-α和IL-1β分泌及其mRNA的表达减轻紫外线辐射引起的皮肤损伤。
Objective To study the protective mechanism of tea polyphenol and aloin on ultraviolet-induced skin damage.Methods The secretions of TNF-αand IL-1βof cultured keratinocytes were detected by enzyme-linked im-munoadsordent assay(ELISA).The mRNA levels of TNF-αand IL-1βwere determined by reverse transcriptase poly-merase chain reaction(RT-PCR).Results The secretions and the mRNA expression of TNF-αand IL-1βwere obvi-ously increased after ultraviolet B(UVB)irradiation comparing with the control group.After treated with tea polyphenol and aloin,the secretions were both decreased.They had significantly difference in statistics(P<0.01).Conclusion Tea polyphenol and aloin treatment can relieve the skin damage induced by UVB through decreasing the secretion s of TNF-αand IL-1βand their mRNA.expression
