5个不同肺癌细胞株的基因差异表达谱 被引量:4

摘要 目的 :希望获得细胞间基因表达的异同 ,对癌细胞的形成和发展中基因参与提供分析线索。方法 :选用 5个不同转移潜能及来源的体外培养肺癌细胞株H12 99,HLAMP ,PAa ,PGCL3和NiS作为研究对象 ,以永生化的支气管上皮细胞 16HBE为对照 ,采用含有 80 0个基因探针的肺癌相关基因芯片LUs,研究了基因表达的差异。结果 :5个肺癌细胞株差异表达的基因共有 35 5个 ,但共同上调或下调的基因仅有 4个 ,基因表达的变化在不同细胞株之间明显表现出多态性。比较三个高转移细胞株和两个低转移细胞株 ,有 12个基因表达改变出现在高转移细胞株而在低转移株未见明显变化 ,其中有 3个已报道的转移相关基因和 9个未报道和转移相关的基因 ,这些基因大多是膜蛋白、细胞骨架蛋白或 /和细胞信号转导有关 ,基因功能复习提示和肿瘤转移有可能的关联性。结论 :cDNA芯片对于肺癌相关基因的筛选是一个非常有效的研究手段 ;对于临床诊断应用 ,需要在大规模数据收集的基础上建立必要的模式 ,通过多基因系统分析 ,才能获得有效的应用。 Objective:To better understand molecular pathology of lung cancer, and provide evidences and data for designing the early diagnosis and therapy protocols.Method:We used 5 cell strains of different sources and metastasis potential to compare the differential expression of 800 genes arrayed in Lus cDNA chips. The lung cancer cell strains used in this experiment were H1299, HLAMP, PAa,PGCL3 and NiS, plus 16HBE as control. Results:The result showed that there were totally 355 genes changed in the comparison of 5 tumor cell strains to the 16HBE cell. Only 4 genes showed the same expression tendency among all 5 strains and the expression profile showed in a diverse way. It was observed that there were 12 genes whose expression were changed in 3 cell strains present high metastasis potential while remained unchanged in the 2 cell strains without metastasis character. Among them 3 were metastasis associate-genes reported by others, other 9 genes had not been reported associate to metastasis in any way. The function of these genes include integral membrane proteins, cell skelecton proteins and the molecules associate to signal transduction. Review of these gene functions and the roles played in the cell hinted possible roles in the metastasis of lung cancer. Conclusions: The cDNA chip method is a powerful tool for exploring the lung cancer associate gene in research. For the clinical applications, patterns of gene expression should be established based on large scale data collection from patients with different kinds and stages of lung cancer.
