摘要
目的 探讨川芎素 (sodiumferulate ,SF)对脑缺血性损伤的保护作用及量效关系 .方法 32只雄性SD大鼠 ,随机分为 4组 ,即对照组、SF 5 0 ,SF 10 0和SF 2 0 0 ,分别在缺血时ip生理盐水 4mL和SF 5 0 ,10 0 ,2 0 0mg·kg-1(SF用 9g·L-1生理盐水溶解 4mL) .采用颈内动脉丝线栓塞致大脑中动脉阻闭 (middlecerebralarteryocclusion ,MCAO)模型 ,术后观察神经行为学变化并于 2 4h时评分后处死动物 ,取大脑行TTC染色以测量脑梗死容积 .结果 2 4h神经功能评分 ,对照组为(1 88± 0 6 4 ) ,明显高于各保护组 (P <0 0 1)SF 5 0 (0 88±0 6 4 ) ,SF 10 0 (0 5 0± 0 5 3) ,SF 2 0 0 (0 6 3± 0 5 2 ) ;梗死容积对照组为 (16 8 1± 4 2 2 )mm3 ,明显大于各保护组 (P <0 0 1)SF 5 0 (79 6± 2 5 8)mm3 ,SF 10 0 (5 8 1± 2 1 6 )mm3 ,SF 2 0 0 (6 4 3± 2 2 6 )mm3 .各保护组间 2 4h神经功能评分和梗死容积无差别 .结论 SF对短暂性局灶性脑缺血损伤呈非剂量依赖性保护作用 .
AIM To investigate the protective effects of sodium ferulate (SF), an intravenous drug made from traditional Chinese herbs, on transient focal cerebral ischemic injury in rats. METHODS 32 male SD rats were divided into 4 groups randomly : Control group (n=8) received no pharmacologic intervention but normal saline 4ml, Group SF 50 ( n =8), SF 100 ( n =8), SF 200 ( n =8) received intraperitoneal SF 50, 100 and 200 mg·kg -1 respectively (SF was dissolved in normal saline 4 mL) just before middle cerebral artery occlusion (MCAO) with 3 0 nylon monofilamen for 120 min. The neurological outcome was evaluated at 24 h after reperfusion. The infarct volume was then assessed after TTC staining following the last neurological outcome evaluation. RESULTS The neurologic deficit score (NDS) of Group SF 50, Group SF 100 and Group SF 200 at 24 h after reperfusion were 0 88±0 64, 0 50±0 53 and 0.63±0 52 respectively, deceasing significantly ( P < 0 01 =compared with those of Control group (1 88±0 64). The infarct volume of Control group (168 1±42 2) mm 3 was significantly greater ( P < 0.01 = than those of group SF 50 (79 6±25 8 )mm 3, SF 100 (58 1±21 6) mm 3 and SF 200 (64 3±22 6) mm 3 at 24 h after reperfusion. There were no differences of NDS and infarct volume among SF pretreated groups. CONCLUSION SF significantly reduces neurologic injury induced by transient MCAO without dose response effect.
