摘要
目的 研究氟对大鼠脑有髓神经髓鞘形态结构及化学组份的影响 ,进一步探讨氟对大脑髓鞘碱性蛋白 ( MBP)及髓鞘磷脂质损伤机理。方法 取饮用含 F- 15 0 m g/ L水 7个月大鼠的脑组织 ,应用 Anti- MBP-HRP免疫组化法标记大脑髓鞘碱性蛋白分布、用组织化学法观察大脑髓鞘脂质改变、用 MBP- EL ISA法分析血清中 MBP含量。结果 免疫组化显示 ,氟中毒大鼠大脑髓鞘染色变浅 ,髓鞘结构松解紊乱和均质化 ,证明髓鞘MPB缺失或溶解。组化法显示 ,氟组大鼠大脑髓鞘磷脂溶解 ,成空泡状或絮状样变 ,且染色变浅 ,说明高氟对髓鞘磷脂产生了破坏作用 ;实验组动物血清中 MBP含量显著升高与对照组比较差异有显著意义 ,个体间 MBP含量高低与免疫组化结果呈相关关系。结论 氟中毒可造成大鼠大脑中枢有髓神经髓鞘直接损伤 ,髓鞘 MBP缺失 ,髓鞘脂质溶解 ,髓鞘结构被破坏。
Objective To study the effect of fluoride (F-) on brain myelin sheath structure and serum myelin basic protein (MBP) content,and to research the pathological mechanism of the injuries to MBP and myelinic phospholipid by F-.Methods Wistar rats were given drinking water containing 150 mg/L F- for seven months.Then the distribution of the rats brain MBP was marked by using MBP-HRP immunohistochemistry (IHC),the change myelinic phospholipid was observed by using solochrome cyanin R·S histochemistry,and the serum MBP content using MBP-ELISA was determined.Results The result of IHC showed the stain of brain MBP of rats with chronic fluorosis was thinner than that of control group,and Lamelliform structure of myelin sheath became relaxing,disorder and homogenization,which demonstrated MBP's release or solution.The result of histochemistry showed myelinic phospholipid solution,vacuolar degenration or cloudy swelling,and the stain became thinner,which demonstrated F- impaired the myelinic phospholipid.The serum MBP content is significantly higher than that of control group (P<0.05).There is a correlation between myelin sheath damage and serum MBP content.Conclusions F- could directly impair myelin sheath to make MBP release,myelinic phospholipid solution and serum MBP content increase.
