纤维蛋白胶载神经生长因子促进周围神经再生的研究 被引量:11

Experimental studies of peripheral nerve regeneration promoted by NGF mixed with fibrin glue as a carrier

摘要 目的 探讨纤维蛋白胶 (FG)作为神经生长因子 (NGF)载体对周围神经再生的促进作用。方法 将Wistar大鼠 96只随机分为 4组 ,即对照组、FG组、NGF组、FG +NGF组 ,每组 2 4只 ,以大鼠左侧坐骨神经为修复神经模型进行实验。术后 2、4、8周进行组织学检查 ,术后 8周进行电生理检查、肌湿重检测、图像分析和透射电镜观察。结果 FG +NGF组的神经传导速度、肌张力、肌湿重、有髓纤维截面积恢复率分别为 ( 84.10± 2 .33) %、( 83 .88± 2 .96 ) %、( 6 2 .5 4± 5 .94) %、( 71.0 1± 3 .6 2 ) % ,吻合口有髓纤维通过率为 ( 6 5 .5 1± 4.5 4) % ,高于对照组、FG组 ( P <0 .0 1) ,高于NGF组 (P <0 .0 5 ) ,FG +NGF组大鼠坐骨神经再生优于单纯NGF组。结论 纤维蛋白胶可作为NGF的载体 ,用纤维蛋白胶载NGF修复周围神经损伤 ,有促进周围神经再生的作用。 Objective To investigate the effects of exogenous nerve growth factor (NGF) mixed with fibrin glue (FG) as a carrier promoting peripheral nerve regeneration.Methods 96 male Wistar rats were randomly divided into four groups:control group,fibrin glue (FG) group,NGF group,and NGF mixed with fibrin glue (NGF+FG) group,with 24 rats in each group.Left sciatic nerves of the rats in each group served as repaired models.Histological examination was made at 2,4,and 8 weeks postoperatively.Electrophysiological examination,muscle wet weight measurement,image analysis,and transmission electron microscopic observation were carried out at 8 weeks postoperatively.Results The recovery rate of nerve conduct velocity,muscle tension,muscle wet weight and myelinated fiber cross section area were(84.10±2.33)%,(83.88±2.96)%,(62.54±5.94)%,(71.01±3.62)% respectively.The cross rate of myelinated fiber at anastomotic stoma was(65.51±4.54)% in the FG+NGF group,higher than in the control group and FG group(P<0.01),higher than in the NGF group(P<0.05).Sciaticnerve regeneration of the rats in the NGF mixed with fibrin glue group was better than in the simple NGF group.Conclusion Fibrin glue can be used as a carrier for NGF to repair injured peripheral nerves and promote peripheral nerve regeneration.
