摘要 〖目的〗探讨血清标本4℃保存7 d对荧光定量PCR检测HBV DNA结果的影响.〖方法〗对来自本院感染科住院患者的52份血清标本进行3次检测:即时检测,4℃保存7 d后检测,-30℃保存1~3个月后统一检测.比较4℃保存7 d后检测结果与其他两次检测的结果,观察其结果的差异有无统计学意义及其符合率.〖结果〗4℃保存7 d后检测结果与即时检测的结果的符合率为62.5%~73.1%,与-30℃保存1~3个月后统一检测结果的符合率为78.1%.4℃保存7 d后检测结果与其他两次结果的比较,其均数差异无统计学意义(t=1.85,t=1.61,P>0.05).〖结论〗以0.6对数单位为允许误差范围,血清标本4℃保存7 d对荧光定量PCR检测HBV DNA结果无影响. Objectives To study the effects of sera storage at 4℃ for 7 days on fluorimetric quantification of HBV-DNA by real-time PCR detection. Methods) HBV-DNA of fifty-two serum specimens from hepatitis B patients admitted in department of infectious diseases of our hospital were detected for three times. The first detections were performed as soon as the blood was drawn. The second and the third detections were done after the blood was stored at 4C for 7 days and at -30C for 1-3 months, respectively. The results of three detections were compared one another statistically. [Results] Coincidence rate between the results of the first and second detection was 62.5%-73.1 %. It was 78.1% between the results of the second and third detection. The difference of the means among results of three detections showed no statistical significance(t=1.85, t =1.69, P >0.05). Conclusion If 0.6 logarithmic unit of error is permitted, it is acceptable method to store the serum specimens at 4C for less than 7 days, thus the fluorimetric quantification of HBV-DNA by PCR detection will not be affected.
