胆囊癌hTERT、P53表达及相关性研究

Expression of hTERT and P53 in carcinoma of gallbladder

目的观察端粒酶催化亚基(hTERT)、P53在胆囊癌中的表达,探讨hTERT和P53与胆囊癌的相关性。方法采用免疫组化法检测42例胆囊癌组织、22例胆囊腺瘤组织、20例慢性胆囊炎组织hTERT及P53的表达。结果慢性胆囊炎组织、胆囊腺瘤组织、胆囊癌组织hTERT阳性表达率分别为0%(0/20)、18.18%(4/22)、80.95%(34/42);P53阳性表达率分别为10%(2/20)、22.73%(5/22)、61.90%(26/42);胆囊癌组织hTERT及P53阳性表达率显著高于慢性胆囊炎组织和胆囊腺瘤组织(P<0.01)。hTERT及P53阳性表达与胆囊癌病理分级和临床分期无相关性(P>0.05)。hTERT和P53在胆囊癌组织中的表达明显正相关(P<0.01)。结论hTERT和P53蛋白过度表达在胆囊癌发生过程中具有重要意义,P53的突变可能与hTERT激活有关。

Objective To explore the expression of human telomerase reverse transcriptase(hTERT) and p53 in carcinoma of gallhladder.Methods Immunohistochemistry was employed to detect the expression of hTERT and P53 in 42 carcinoma of gallbladder、22 adenoma of gallbladder and 20 chronic cholecystitis.Results The positive expression rates of hTERT were 0(0/20),18.18%(4/22) and 80.95%(34/42),and the positive expression rates of P53 were 10%(2/20),22.73%(5/22) and 61.90%(26/42),in chronic cholecystitis,adenoma of gallbladder and carcinoma of gallbladder respectively.The positive expression rates of hTERT and p53 in carcinoma of gallbladder were significantly higher than those in chronic cholecystitis and adenoma of gallbladder(P<0. 01).The positive expression of hTERT and p53 in carcinoma of gallbladder were not related with the histologic grade and clinical stage(P>0.05).The expression of hTERT was a positive correlation with p53 expression in carcinoma of gallbladder(P<0.01).Conclusion The expression of hTERT and p53 is related to the occurrence of carcinoma of gallbladder.Overexpression of p53 may play a role in the transcriptive activation of hTERT in carcinoma of gallbladder.
