蛋白酪氨酸磷酸酶Shp2与接头蛋白Gab2在子宫内膜癌中的表达及意义 被引量:1

摘要 目的探讨蛋白酪氨酸磷酸酶Shp2与接头蛋白Gab2在子宫内膜癌中的表达、两者的相关性及其与预后的关系。方法采用免疫组化SP法,检测50例子宫内膜癌(癌组)、50例子宫内膜不典型增生(癌前病变组)及30例正常增殖期子宫内膜(正常组)中Shp2蛋白和Gab2蛋白的表达水平,分析其表达与子宫内膜癌临床病理参数的关系,及二者的相关性。结果正常组、癌前病变组和癌组的子宫内膜组织中,Shp2蛋白的阳性表达率分别为0、12.00%、64.00%,Gab2蛋白的阳性表达率分别为3.33%、38.00%、58.00%,且组间比较均有统计学意义(P<0.05);Shp2蛋白及Gab2蛋白的阳性表达率在肌层浸润深度及有无淋巴结转移组中比较,差异均有统计学意义(P<0.05);多因素生存分析显示:Shp2蛋白阳性表达者的总生存率明显下降,Gab2蛋白阳性表达者总生存率也明显下降;子宫内膜癌中Shp2蛋白的表达与Gab2蛋白的表达呈正相关(r=0.368,P<0.05)。结论 Shp2蛋白及Gab2蛋白的表达可能参与了子宫内膜癌的发生、发展、浸润等过程,有可能作为判断子宫内膜癌预后的新指标。 Objective To investigate the expression of protein tyrosine phosphatase 2(Shp2) and the adapter protein Gab2 in endometrial carcinoma, and to analyze the correlation between the two proteins as well as that between their expression and prognosis. Methods The expression levels of Shp2 protein and Gab2 protein were detected by immunohistochemical SP method in 50 patients of endometrial carcinoma(cancer group), 50 patients of endometrial atypical hyperplasia(precancerous lesion group) and 30 subjects of normal proliferative phase endometrium(normal group).The correlation between the expression and the clinicopathological parameters of endometrial carcinoma were analyzed,as well as the correlation between the two proteins. Results The positive expression rate of Shp2 protein was 0.00%,12.00%, 64.00% in normal group, precancerous lesion group, cancer group, respectively, and the positive expression of Gab2 protein was 3.33%, 38.00%, 58.00%, respectively, with statistically significant difference between the three groups(P<0.05). The positive expression rate of Shp2 protein and Gab2 protein showed no statistically significant difference between groups of different muscle layer infiltration depth and the groups with or without lymph node metastasis(P<0.05).Multivariate survival analysis showed that the total survival rate of patients with positive expression of Shp2 protein was decreased significantly, and the total survival rate of patients with positive expression of Gab2 protein was also significantly decreased. The expression of Shp2 protein was positively correlated with the expression of Gab2 protein in endometrial carcinoma(r=0.368, P<0.05). Conclusion The expression of Shp2 protein and Gab2 protein may be involved in the occurrence, development and infiltration process of endometrial carcinoma, what can be used as new prognostic indicators for evaluating the prognosis of endometrial carcinoma.
