果王素对肺癌GP方案化疗骨髓抑制小鼠的保护作用 被引量:2

Effect of Kiwi essence in the prevention and treatment of bone marrow inhibition in mouse models of lung cancer undergoing GP chemotherapy regimen

摘要 目的观察果王素对肺腺癌小鼠使用GP方案化疗过程中对骨髓抑制的保护作用,初步探讨果王素防治骨髓抑制的机制。方法 32只接种Lewis肺癌的C57BL/6J小鼠随机分成对照组、化疗组(GP方案)、低剂量果王素联合化疗组及高剂量果王素联合化疗组。每组8只,接种Lewis肺癌第四天开始予以相应药物干预,于接种后24 d处死各组小鼠,经眼球静脉丛取各组小鼠血液样本2 ml,行血常规检测,酶联免疫吸附法(ELISA)测定血清单核细胞趋化蛋白1(MCP-1)及白介素-2(IL-2)的浓度,取股骨计数骨髓有核细胞数,并测定超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)、总抗氧化能力(T-AOC)的含量,取胸腺称重,计算胸腺指数。结果四组小鼠的RBC计数组间两两比较差异均无统计学意义(P>0.05);低剂量果王素联合化疗组、高剂量果王素联合化疗组与化疗组、对照组比较在WBC、血单核细胞(BMNC)、胸腺指数、SOD、T-AOC方面差异均有统计学意义(P<0.05),低剂量果王素联合化疗组、高剂量果王素联合化疗组与化疗组比较WBC、PLT、BMNC、胸腺指数、SOD、T-AOC、IL-2明显升高(P<0.05),低剂量果王素联合化疗组、高剂量果王素联合化疗组与化疗组比较MPO、MCP-1明显下降,差异均有统计学意义(P<0.05)。结论果王素能明显减轻化疗中骨髓抑制的程度,其可能机制与增强免疫、清除氧自由基、抑制氧化应激及抑制炎症反应有关。 Objective To investigate the protective effect of Kiwi essence for bone marrow inhibition in mouse models of lung cancer undergoing GP chemotherapy regimen, and to explore the related mechanisms. Methods Thirty-two C57BL/6J mice bearing Lewis cells were randomized into control group, chemotherapy group, low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group, with 8 mice in every group. The mice were given corresponding drug intervention on the 4thday after establishment of models, and then sacrificed on the 24 thday. Blood samples(2 ml) were collected from eye venous plexus in each group for routine blood test and enzyme-linked immunosorbent assay(ELISA), with the serum levels of monocyte chemotactic protein-1(MCP-1)and interleukin-2(IL-2) determined. Femur samples were collected for bone marrow nucleated cell counts and determination of the levels of myeloperoxidase(MPO), superoxide dismutase(SOD), and total antioxidant capacity(T-AOC).Thymus gland was taken to determine the weight of thymus and thymus index. Results There was no statistically significant difference between the four groups in WBC(P>0.05). Compared with control group and chemotherapy group,WBC, blood mononuclear cell(BMNC), thymus index, SOD and T-AOC in low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group showed statistically significant difference(P<0.05). Compared with chemotherapy group, WBC, blood platelet(PLT), BMNC, thymus index, SOD, T-AOC and IL-2in low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group were significantly increased(P<0.05), while MPO and MCP-1 were significantly decreased(P<0.05). Conclusion Kiwi essence can significantly reduce the degree of bone marrow inhibition in chemotherapy. The possible mechanism may be associated to its effect of enhancing immune function, removing oxygen free radicals, inhibiting oxidative stress and inhibiting inflammatory reaction.
