摘要
哺乳动物Sirtuins家族是酵母沉默信息调节因子2(SIR2)的同源蛋白质的统称,属于一类组蛋白/非组蛋白去乙酰化酶,组成成分为SIRT1~SIRT7,依赖主体为烟酰胺腺嘌呤二核苷酸(NAD+)。Sirtuins家族可作用一系列底物,参与多种细胞生物学功能,对细胞的生存、衰老、凋亡等生理活动有着重要的调节作用,并且与代谢综合征、肿瘤、心血管疾病、神经退行性疾病等疾病的发生密切相关,其中,沉默调节蛋白1(SIRT1)与SIR2的同源性最高,也是近年来Sirtuins家族成员中得到最深入研究的一项。本研究现就SIRT1与代谢相关疾病的研究进展作如下综述。
Sirtuins family, a general term for the homologic protein of silent information regulator 2(SIR2) of yeast, belongs to a class of mammalian histone/non-histone deacetylase, which is composed of SIRT1~SIRT7 and relies on nicotinamide adenine dinucleotide(NAD+). Sirtuins family can serve as various substrates and participates in various cellular biological functions, modulating cellular survival, aging, and apoptosis. It has been reported that sirtuins may be associated with the occurrence of metabolic syndrome, tumors, cardiovascular disease, and neurodegenerative disease.Among the Sirtuins family members, SIRT1 shares the highest homology with SIR2, and it has obtained the most in-depth study. The study reviews the research progress of SIRT1 and metabolic diseases.
