摘要
目的探讨骨肉瘤患者血清基质金属蛋白酶-9(MMP-9)和Ki-67的表达及与肿瘤侵袭、转移、复发的相关性。方法选取2011年1月至2014年5月本院骨科收治骨肉瘤患者65例,同期纳入50例体检健康者为健康对照组;比较骨肉瘤患者和对照组患者的血清MMP-9和Ki-67水平;分析血清MMP-9和Ki-67的水平与骨肉瘤临床病理特征的相关性;比较随访期间未复发与复发患者间血清MMP-9和Ki-67水平。结果骨肉瘤患者血清MMP-9和Ki-67水平分别为(2.78±0.29)μg/ml和(52.33±5.46)ng/ml,均显著高于对照组[(1.31±0.16)μg/ml、(20.57±2.39)ng/ml](P<0.01);骨肉瘤患者血清MMP-9和Ki-67水平与其年龄、性别、瘤灶大小、肿瘤部位及Dahlin's分型无显著相关性(P>0.05),而与GTM分期及转移存在显著相关(P<0.01);1年期随访结果显示,35例复发,30例未复发;复发组患者入院时血清MMP-9和Ki-67的水平分别为(2.93±0.34)μg/ml和(54.73±5.86)ng/ml,明显高于非复发组同期水平[(2.72±0.32)μg/ml、(50.12±5.35)ng/ml],差异均有显著统计学意义(P<0.01)。结论骨肉瘤患者的血清MMP-9和Ki-67水平显著增高,与骨肉瘤的侵袭、转移、复发密切相关,是临床评价该病患者治疗效果的重要指标。
Objective To investigate serum levels of matrix metalloproteinases 9(MMP-9) and Ki-67, and its correlation with the invasion, metastasis and recurrence of osteosarcoma. Methods Sixty-five patients of osteosarcoma were chosen in our hospital from January 2011 to May 2014. During the same period, 50 patients of routine physical examination in our hospital were enrolled as the control group. Serum levels of MMP-9 and Ki-67 between two groups were compared. Correlation of serum levels of MMP-9 and Ki-67 and clinicopathological features of osteosarcoma was analyzed. Serum levels of MMP-9 and Ki-67 in the patients with and without recurrence group were compared. Results Compared with control group, serum levels of MMP-9 and Ki-67 in osteosarcoma patients were(2.78±0.29) μg/ml and(52.33±5.46) ng/ml respectively, which were significantly higher than those in the control group [(1.31±0.16) μg/ml,(20.57±2.39) ng/ml](P<0.01). Serum levels of MMP-9 and Ki-67 of osteosarcoma patients were irrelevant to age, gender, the size of tumor, tumor site and type of Dahlin’s(P>0.05), and were relevant to GTM grade and metastasis(P<0.01). All patients were followed up for one year with 35 cases of recurrence and 30 cases without recurrence. Serum levels of MMP-9 and Ki-67 of the recurrence group were(2.93 ± 0.34) μ g/ml and(54.73 ±5.86) ng/ml, which were significantly higher than those in non-recurrence group [(2.72±0.32) μg/ml,(50.12±5.35) ng/ml](P<0.01). Conclusion Serum levels of MMP-9 and Ki-67 of osteosarcoma patients are significantly increased, and they are closely related with invasion, metastasis and recurrence of osteosarcoma, which is an important indicator for evaluating the clinical therapeutic effect.
