摘要
目的探讨血清和糖皮质激素诱导激酶1(SGK1)rs1743967以及血清和糖皮质激素诱导激酶2(SGK2)rs743998与自然流产之间的关系。方法采用病例对照研究方法,选取2013年1~12月我院64例自然流产患者作为自然流产组,同期40例人工流产患者作为对照组,利用3730XL测序仪检测SGK1 rs1743967和SGK2rs743998基因多态性,χ2检验比较病例组与对照组基因型及等位基因频率分布。结果 1 SGK1 rs1743967对照组基因型为GG+GT,而自然流产组以GG+GT为主,其次是TT,两组基因型比较差异有统计学意义(P<0.05)。对照组等位基因G和T频率各占50.0%;自然流产组G占35.2%,T占64.8%,两组等位基因频率比较差异有统计学意义(P<0.05)。2 SGK2 rs743998对照组基因型以CT+TT为主,其次是CC,而自然流产组以CC为主,其次是CT+TT,两组基因型比较差异有统计学意义(P<0.05)。对照组等位基因C和T频率各占50.0%;自然流产组C占83.6%,T占16.4%。两组等位基因频率比较差异有统计学意义(P<0.05)。结论 SGK1 rs1743967和SGK2 rs743998与自然流产有关,但相关机制有待今后进一步研究。
Objective To investigate the association of serum and glucocorticoid-regulated protein kinases 1(SGK1)and SGK2 polymorphisms(rs1743967 for SGK1 and rs743998 for SGK2) with spontaneous abortion. Methods Sixty-four cases of spontaneous abortion, who enrolled into our hospital from January 2013 to December 2013, were selected as the spontaneous abortion group, and 40 cases of artificial abortion were selected as a control group for the same period.Polymorphisms of rs1743967 and rs743998 were determined through 3730 XL sequencer. genotype and allele frequency distribution of the case group and the control group were compared with χ2test. Results 1 rs1743967 genotype of SGK1 in the control group were GG+GT, while rs1743967 genotype of SGK1 in the spontaneous abortion group were mainly GG+GT, followed by TT. Control group and spontaneous abortion group had a statistically significant difference(P<0.05). Allele frequency of G and T in the control group was equally 50.0%, while allele frequency of G and T in spontaneous abortion group were respectively 35.2% and 64.8%, and allele frequency distribution has a statistically significant difference between control group and spontaneous abortion group(P<0.05). 2 rs743998 genotype of SGK2 in control group was mainly CT+TT, followed by the CC, while spontaneous abortion group was mainly CC, followed by CT+TT, and genotype of the two groups have a statistically significant difference(P<0.05). Allele frequency of C and T in control group was equally 50.0%,while allele frequency of C and T in spontaneous abortion group was respectively 83.6% and 16.4%. Allele frequency distribution of rs743998 had a statistically significant difference between control group and spontaneous abortion group(P<0.05).Conclusion SGK1 and SGK2 polymorphisms(rs1743967 for SGK1 and rs743998 for SGK2) have the correlation with spontaneous abortion. However, the mechanisms have yet to be studied in detail.
